A theoretical drug is being studied for pharmacokinetic properties. It has a clearance of 10 L/min and an elimination constant of 0.2 min-1 . Which of the following is the best estimate for volume of distribution of this drug?

a) 2 liters

b) 20 liters

c) 50 liters

d) 100 liters

e) 50 mL

Answer

Answer c.

Based on the formula Ke = CL / Vd, the Vd may be solved for as Vd = CL / Ke. Thus, the volume of distribution of this drug is CL / Ke = 10 L/min / 0.2 min-1 = 50 Liters. Note that it is very important to be consistent with units of measure (mL vs. L, min vs. hour) for these calculations.
